Ordered rabbit antiserum against a peptide of interest. Tested pre-immune serum of 4 rabbits to pick 2 with low background in the size of the protein of interest using Western blot. All had already multiple bands, so we tried 4 more with same result. We then got 4 more and only one looked OK, so we started with the immunization. Once we received post-immune bleeds, we detected a faint band for the protein of interest and a lot of unexplained background bands appeared.
